Breakthrough Discovery Links Brain Immune Cells to Alzheimer's Sleep Loss

Scientists have uncovered a surprising culprit behind sleep disruption in Alzheimer's disease: the brain's own immune cells, not the amyloid plaques previously blamed.

Researchers at the University of Kentucky's Sanders-Brown Center on Aging found that overactive microglia—the brain's resident immune cells—are the primary cause of sleep loss. When they used a drug to temporarily silence 87% of these cells in mice with Alzheimer's pathology, the animals gained more than two hours of restorative sleep per night, without removing a single plaque.

The discovery, published in the journal Alzheimer's & Dementia, has immediate implications for treatment. Researchers are now exploring whether existing medications, such as the diabetes drug metformin, could reset how these immune cells behave.

By targeting this process, scientists hope to restore sleep and improve quality of life for patients years before memory loss begins, potentially helping with attention, cognition and confusion.
